Xencor (XNCR) Q3 2026 earnings review
Xencor extends its cash runway, while trial costs climb fast
Xencor brought in cash from partners and legal settlements while spending much more on its own clinical trials. The biotechnology developer of engineered antibodies posted $117 million in revenue and a $39 million net profit after collecting a royalty settlement from Alexion and a Phase 3 milestone from Astellas. Chief executive Bassil Dahiyat selected a late-stage dose for lead kidney cancer drug XmAb819 and raised the year-end cash outlook by $60 million, though by our math the plan points to $66 million of cash use in the fourth quarter.
| Revenue | $116.5M $21.0M a year ago, lifted by settlements and milestones |
|---|---|
| Research and development spending | $84.3M +55% from a year ago |
| Lead kidney cancer drug response rate | 45.5% 5 of 11 patients at selected Phase 3 dose |
| Year-end 2026 cash outlook | Raised $60M $480–500M range; funds operations into 2029 |
⚖️ Verdict: 🟢 Bullish
The story got better because the kidney cancer drug cleared its dose-selection hurdle and non-dilutive cash extended the funding runway into 2029. Selecting the 400 microgram dose with a 46% confirmed response rate lets Xencor seek accelerated approval talks with regulators in early 2027. One caution: research spending jumped 55% from a year ago to $84 million, and the company quietly dropped standalone late-stage plans for its ovarian cancer candidate XmAb541.
The question now is whether the 46% tumor response rate for XmAb819 holds up beyond the first 11 evaluable patients. If responses stay high as the rest of the expansion group matures, the drug could qualify for an accelerated regulatory filing; if the rate drops in a larger sample, Xencor will have to wait for a full Phase 3 trial. Updated data from the cohort in the first half of 2027 will tell.
🐂 Bull Case
Stock Sales and Settlements Extend Cash Into 2029
Xencor, a biotechnology company developing engineered antibodies for cancer and autoimmune diseases, raised its year-end cash target by $60 million at the middle of the range. Management now expects to finish 2026 with $480 million to $500 million, enough to "fund research and development programs and operations into 2029" instead of through 2028.
- Equity sale proceeds: $66.9 million in cash from selling stock in a partner
- Alexion settlement cash: $52.5 million received in August, with another $52.5 million due in the third quarter of 2027
- Partner milestones: $25.0 million triggered by Astellas and $2.5 million by Zenas BioPharma
These inflows lifted total cash and marketable debt securities by $69.3 million during the quarter to $555.7 million. That extra cushion matters because the drug developer can now run its planned late-stage kidney cancer trial and its Phase 2 ulcerative colitis study through their 2027 readouts without needing to sell its own shares.
What to watch: the collection of the final $52.5 million Alexion payment in the third quarter of 2027. Receiving that installment on schedule will keep the cash buffer above $400 million as late-stage trial costs peak.
Lead Kidney Cancer Drug Picks a Phase 3 Dose
"Xencor’s lead CD3 T-cell engaging bispecific antibody, XmAb819, has demonstrated compelling clinical data in advanced clear cell renal cell carcinoma," chief executive Bassil Dahiyat said. XmAb819 is an antibody designed to link a patient's immune T-cells directly to kidney tumors that carry the ENPP3 protein.
In October, the company presented Phase 1 results and selected 400 micrograms per kilogram as its target intravenous dose for late-stage testing. Among 11 evaluable patients at that dose, five had confirmed tumor shrinkage (a 45.5% response rate) and one more had an unconfirmed response, while 5.9% of all 68 expansion patients stopped treatment due to related side effects.
Selecting a dose clears the way for late-stage trials. Dahiyat plans to ask the Food and Drug Administration in the first half of 2027 about an accelerated approval path for "subsets of patients with advanced ccRCC who have exhausted standard-of-care treatment options", and to start a Phase 3 trial against the kidney cancer drug tivozanib by year-end 2027.
What to watch: updated results from additional patients enrolled in the 400 microgram intravenous cohort in the first half of 2027. Keeping the confirmed response rate near 45% in a larger group would strengthen the case for accelerated approval.
Partner Trials Trigger $27.5 Million in Milestones
Before Xencor's own drugs reach the market, it earns revenue when pharmaceutical partners advance antibodies built with its XmAb engineering technology. Dahiyat noted that "by the end of 2027 we anticipate XmAb819 will be the third XmAb 2+1 bispecific antibody to enter a potential registration-enabling study, following xaluritamig and ASP2138".
All three major late-stage partner programs hit milestones this quarter. Astellas dosed the first stomach cancer patient in a Phase 3 trial of ASP2138, triggering a $25.0 million payment, while Zenas partner Bristol Myers Squibb filed for Japanese approval of the autoimmune drug obexelimab, triggering $2.5 million. Amgen also finished enrolling its first Phase 3 prostate cancer trial for xaluritamig.
What to watch: the Food and Drug Administration review of Zenas BioPharma's application for obexelimab, which the agency accepted in August 2026. An approval in 2027 would unlock commercial milestones and royalties ranging from the mid-single digits to the mid-teens.
🐻 Bear Case
Research Spending Accelerated 55% as Trials Expanded
Xencor reported $39.4 million in net income and $22.9 million in operating profit, turning its operating margin positive by 245.9 percentage points from a year ago. However, one-time revenue items drove that profit, masking a steep rise in the cost of running its clinical trials.
- Research and development spending: $84.3 million, up 55.1% from a year ago and up 17.2% from last quarter
- One-time revenue items: $27.5 million in partner milestones plus the scheduled $77.4 million Alexion royalty settlement recognition
- Underlying operating loss: about $82.0 million by our math after stripping out those settlement and milestone items
Because revenue quadrupled on those one-off items, research and development fell by 186.6 percentage points as a share of revenue. In dollar terms, though,
"higher external and internal costs associated with development programs" pushed quarterly operating expenses to $93.7 million. By our math, stripping out the equity sale, the Alexion cash installment and the two milestones leaves an underlying cash burn of about $77.6 million in Q3 and $65.7 million implied for Q4.
What to watch: quarterly research and development expenses as XmAb819 prepares for Phase 3 in 2027. If trial spending stays above $85 million a quarter without new partner payouts, the cash buffer will shrink faster than the 2029 plan assumes.
Ovarian Cancer Drug Drops Late-Stage Target
Earlier this year, Xencor planned to present Phase 1 data for its ovarian and testicular cancer antibody XmAb541 in the second half of 2026 to pick a Phase 3 dose. After observing reversible hearing loss at higher doses last quarter, management shifted focus to testing XmAb541 in combination with a second antibody, XmAb808.
In this quarter's pipeline update, the biotechnology group removed standalone XmAb541 from its overview and only expects the combination trial to reach its target dose range in 2027. Dropping the standalone path removes Xencor's second near-term late-stage cancer candidate and leaves oncology upside reliant on XmAb819.
What to watch: whether the combination of XmAb541 and XmAb808 reaches its target dose in 2027 without triggering the same hearing side effects. Clean safety at the target dose is required before Xencor can restart late-stage planning for the program.
👓 Other Themes
Autoimmune Programs Remain on Track for 2027 Readouts
Xencor kept the timelines unchanged across its four autoimmune antibody trials. The company will present early Phase 1 rheumatoid arthritis data for plamotamab on November 9, followed by an update on XmAb657 before year-end. For inflammatory bowel disease, XmAb942 remains on schedule for a blinded interim check around year-end 2026 and full Phase 2b ulcerative colitis results in the second half of 2027, while XmAb412 will report healthy volunteer data in the first half of 2027.
One-Time Legal Recovery Cut Overhead Expenses
General and administrative expenses — the overhead cost of running the company — dropped 33.9% from a year ago to $9.4 million, falling by 59.4 percentage points as a share of revenue. Xencor said the decline came from "a non-recurring recovery of legal expenses" after settling its Ultomiris patent and royalty dispute with Alexion last quarter, so overhead spending should return toward its earlier $16 million to $17 million quarterly pace.
💲 Other KPIs
Liabilities related to the sales of future royalties — upfront cash Xencor raised in prior years to be repaid from partner royalty streams — fell 9.4% from last quarter and 20.6% from the end of 2025. As royalty revenue comes in from partners, the company pays down this non-cash balance without drawing on its cash reserves.
Other income fell from $41.5 million a year ago because of smaller paper gains on partner stock, after swinging from a $50.8 million loss in Q1 and a $15.1 million gain in Q2. Xencor sold $66.9 million of stock during the quarter, cutting its stake in partner Zenas BioPharma by 67.7% from a year ago to 1.0 million shares.
Diluted shares rose 5.3% from a year ago and 3.3% from last quarter. Basic shares grew only 2.4% over the past year to 76.2 million, because employee stock options only count toward the diluted total in quarters when the company reports a positive net profit.
🔮 Guidance
Raised. The drug developer lifted the middle of its year-end cash plan by $60 million, leaving the balance about 20% below the end of 2025. By our math, dropping from $555.7 million in Q3 to the $490 million middle of the range implies about $65.7 million of cash use in Q4. It is the second straight raise after an early cut across four quarters: -$25M, +$40M, +$60M.
Raised. Management now expects its cash reserves to fund research programs and operations into 2029, extending the timeline from its prior target of through 2028. The longer runway rests on the $66.9 million stock sale, the $52.5 million Alexion settlement installment received in August, and $27.5 million in partner milestones.
❓ Key Questions
What was the response rate for all 400 mcg/kg patients?
Xencor reported a 45.5% confirmed response rate in 11 evaluable kidney cancer patients at the 400 microgram dose, but noted that additional patients also enrolled in that cohort. Investors need to know how many total patients received that dose and when their results mature.
How high will quarterly R&D spending climb in 2027?
Research and development expenses jumped to $84.3 million in Q3 even before the planned Phase 3 kidney cancer trial begins in late 2027. Management has not said whether new trial starts will push quarterly spending above $90 million next year.
Will Xencor sell its remaining 1.0 million Zenas shares?
The company raised $66.9 million by selling equity securities this quarter and now holds 1.0 million shares of Zenas BioPharma, down from 3.1 million a year ago. Whether management plans to cash in the rest affects how much non-dilutive funding remains.
How much did the legal recovery reduce Q3 overhead costs?
General and administrative spending dropped by $7.0 million from last quarter to $9.4 million due to a one-time recovery of legal expenses. Disclosing the exact dollar size of that credit would clarify the company's ongoing overhead run rate.
